Distorted marker in SDS - (Apr/27/2007 )

I have been doing SDS of protein samples at 8% however each time there is some problem in marker lane. The band gets totally distorted. However the samples showare perfect bands. Any suggestions

Usually the bands are distored due to high salt content. Check if that is the case.

Thanks! but we have been using milliQ water for gel preparation and for other chemicals and this distortion is only in marker lane not in samples?

hello,

quite often i get a distorted marker , where it kind of 'smiles' either up or down, and just doesnt seperate properly.
i have found (and have been advised) that this quite often happens when you run anything in the outside wells of your gel. i have been told to keep these lanes free if you can help it, and it seems to work!
