Sequencing PCR products with biotin-labelled primer - (Apr/24/2007 )

Hi!

Has anyone sequenced a PCR-product that has a biotinylated primer, with the same biotinylated primer as sequencing primer? Are there usually any problems with this? I would like to cut out a Pyrosequencing PCR-product from the gel and sequence it...

I would be worried (as perhaps you are) that the biotin would change the migration of the ssDNA sequencing product in the capillary. Why not simply make a non-biotinylated primer and use it for sequencing?
