Can anybody help me to know-how I can separate the antigen presenting cell from spleen of rat.

If by antigen presenting cells you mean only macs and dcs and not B cells then you can do a simple adherence step using plates. This allows antigen presenting cells to adhere to the plate and B cells, NK cells, and T cells to remain in the media. There are also several kits you can use to isolate macs and dcs through negative selection that are rat specific.
