DNA Extraction-pigment contamination - (Apr/14/2007 )

I extracted DNA from plant shoot meristem tissue. However, the DNA that I got is reddish brown in color (anthocyanin contamination?) I used the CTAB method and repeated the wash buffer step several times to get rid of the color...didn't work :-(

Can anyone suggest some way to get rid of the color?

Why do you care? Often colored material can be removed with carbon black followed by filtration.

Wouldn't it affect PCR and Southern hybridisation?

Probably not. Try it and see.

Is there a control reaction which is working? Do you know the primers and annealing temperature are working? Often the problem is too much template DNA. How much are you adding?
