Is My Serum Still OK To Use? - (Apr/06/2007 )
I am using Charcoal Stripped Serum for an experiment.
I left a 50 mL tube of CSS in a 37 Celcius water bath for 4 hours. I had originally meant to thaw it and then add it to my alpha MEM media, but I got sidetracked and left it in the water bath.
When I got back, I added the CSS to my alpha MEM media. I don't have much to spare, or otherwise I would have used a new one.
Do you think my serum was OK?
I am trying to grow my cells to confluence for an experiment.

should be fine. Our technician has serum in the water bath for hours before using it.

should be fine. Our technician has serum in the water bath for hours before using it.
May be it succeed for your cells but it is not good, because serum contain short half-life cytokines and activity may decreased. Other cells may be more sensitive for them.
Like a rule I aliqote 500ml bottle in accord with needs ( 15 or 50 ml falcons) and freeze. The day before preparation ( evening before next day) of serum containing media put falcon with serum at 4C allowing mild conditions of thawing . Next day you have ready to use serum. You could add it without heating till 37C) in prewarmed media.
