i have a question"
if i spread 2 plates *BL21 E coli conatining my plasmid* and put them in the refrigerator for one day can i put them after that into 37 degree ? and the same for ON 10 ml culture?or being in 4 degree for a whole day will affect its growth?

It should work fine.

If you want them to grow slowlier (i.g. over the weekend), you can just leave them at room temperature instead of incubating @37°C. I put my plates in the drawer when I do a transformation on Friday and then pick colonies/put them into the fridge Sunday night or Monday morning...
