PCR of methylated DNA? - (Mar/26/2007 )

Hi all,

Does anyone know if you can you use methylated DNA as a PCR template? I would like to check for the presence of an insert in a plasmid isolated from Staphylococcus, and I know restriction digests aren't an option, but hoping PCR will be without further purification. I didn't have much luck last time I tried. Thoughts?

Only some enzymes are methylation sensitive. Methylation will not adversely affect PCR either.
