IPTG protein expression in BL21 (or orthers) - how to measure the expression (Mar/14/2007 )

I need to measure the protein expression before and after IPTG induction. I have i pool og 96 clones and have to measure the difference between the expression of the clones before and after IPTG expression. How to do this?

Seeing as how you have so many clones to test, are you able to do a dot blot or ELISA? That is, are there antibodies available against your protein? You'll need to lyse the cells and extract total protein, then either run an ELISA or blot on a membrane.

If you want to see things on a gel, you might get away with pooling aliquots of groups of clones and running an SDS-PAGE to see if any give you the protein (you could make up 10 pools so you can check all 96 samples on 1 gel). Then you select the successful pool, and test the individual members on SDS-PAGE. A few gels, to be sure, but fewer gels than if you run 96 separate samples (with controls...).
