How can I isolate leucocytes from bone marrow.

cut the 2 edges of the bone, flush the inside with medium (with a needle and seringue), lyse erythrocytes, and wash the cells in medium

Thanks. Supposedly I want to seperate the lymphocytes and the granulocytes from bone marrow samples, can I follow the ficoll procedure or some other precautions needs to be taken.

sorry, I have not read your question correctly
I have no experience with Ficoll separation on BM extracts, but I suppose that there is no reason for special precautions...

As far as I'm aware, ficoll is flexible regarding the source of the cells. The most important parameter is the density of the cells.
