YEAST EXPRESSION - (Mar/04/2007 )

I am using the k. Lactis yeast expression kit for protein expression. I transformed the yeast an d incubated at 28 degree. The protocol says that the colonies seen in 3-4 days. But i can see the colony after the 40 hrs of incubation. I suspect if this is a bacterial colony. But then the growth was not seen last 30 hrs. SO if bacteria grows it grows in 12 hors. If anybody has used the kit before please let me know the colony i have on plates is yeast but not the bacteria.

You can add ampicillin to your yeast medium to inhibit bacterial growth. But the colony color and morphology should tell you if you have yeast or bacteria.
