A. R. grade isopropanol - (Mar/02/2007 )

Dear all,
what is the recommended grade for the reagents use in RNA extraction?
Is molecular grade is a must?

What will happen is i use A.R. grade isopropanol for RNA extractio?
Will it affect the yield? purity? or not working at all?

Thanks
Oche

AR grade is meant to use in analytical chemistry.
Molecular work preferably use molecular grade reagents, because is DNase RNase free.

I am not sure of other effect

As Hadrian said Molecular with molecular grade, but also if you are extracting RNA is better to have reagents for RNA use only. If too many people uses the same reagents for DNA and for RNA without precaution a contamination could result. I always mantein all reagents and materials for RNA apart and don't let any one touch them unless they will work with RNA (and make sure that they know GLP's)...maybe I'm a little manic wacko.gif , but RNA degrate more easily than DNA and I don't want to loose samples and then loosing time doing again an experiment.

However, It seem to me that very little molecular grade reagents are available?
Even Sigma people told me that they have been selling A.R grade to customers for long time and dont get any complain.

So, is molecular grade ethanol and isopropanol essential?

can't really comment on RNA work.

but for DNA work there is no actual difference. More likely your sample is even dirtier then the chemical.
