Double staining antibody plus Annexin! - Help (Feb/26/2007 )

Hey!
I would like to show by double staining that when the gene I'm working with is sillenced the cells do not go to apoptosis. We have already really good data about that, and this experiment is something extra for the paper. Anyway if I will put y siRNAs and then stain with antibody to show decreased in expression level, is it possible to use also Annexin-V as a apoptosis marker. I'm not sure about that. One marker is on the surface of the cells, and then you need to open cells in order to stain with antibody?. Any suggestions. Thank you and wish you have a nice day.

I dont know about annexin but caspase 3 makes for a reliable apoptotic marker

annexin V should be labeld f.i. with Alexafluor 488; do you need stain for flow cytometry or fluorescence microscopy? anyway, annexin V and propidium iodide should be given to living cells
