what more i do?(sequencing) - sequencing (Feb/20/2007 )

Hello

i am in problem of my last stages of work.
my work is actually to sequence the growth corelated genes of chickens.uptil now i have done

rna isolatin(not big work)
cDNA synthesis (not big work)
PCR
gell purification.

and now i am about to send the purified amplified product for sequencing


unsure.gif mellow.gif sad.gif but problem is that sad.gif i want to add some more technique or u can say to elaborate my work .

here i want suggestions what more i can do or add in this line of work

Hi chicken,

do you use rt-pcr? or are you just interested in the sequence? If not, you could use your cDNA for realt-time, maybe you have different time point during development or something like that (treatment, etc.) - then you can compare the expression of your genes. Or, you want to look at the regulation of those genes (promoter construct, DNA methylation?)
So many ideas, I could write the whole day, but basically, you should provide some more information about your experiments ... then these ideas may be more focussed and helpful.

but meaning of my problem is that i have sent amplified genes for sequencing and i have 1 more month i want to do some more work in same line.

my sir has suggested that now i should work on different breeds like i have done on broiler and next i should work on domestic chicken. IS this a good idea? unsure.gif

or any other u have?

well what do you want to learn about your gene?

Are you interested in its activity? Its evolutionary history? How it has change since chickens have been domensticated? How it varies between chicken breed for different purposes? How many gene are there? What do these genes do? Do they belong to any gene families? Any resemblence to human genes/mice gene/mamalian/fish genes? Can I make a super large chicken able to fight Godzilla?

Ask yourself what you want to know about these genes and you will be able to decide what to do next.

after you get the sequence of the gene, you could do a lot more work , like trying to compare it to other studies in other types of chicken or compare it to other animals like mouse or rat.
