Thymus DNA extraction - (Feb/14/2007 )

hello all,
Do you think it's absolutly necessary to lyse the red blood cells if you want to extract DNA from mouse thymus tissue ?
(considering than there no DNA in the red cells)

Hi Francis,

don't think so, you can extract DNA from thymus using standard PC extraction. Maybe you have to do it twice, as thymus (like liver) is very rich in nucleic acids.

Hello, thanks for your reply. Sorry but what do you mean by PC extraction (phenol/chloroform?).
Hum, you had no idea about further potential "PCR problem" due to the erythrocytes ?

Yes, I meant phenol/Chloroform. I heard of this problem, but only for splen... I used rat liver and did not have that problem, and wasn't aware that thymus contains more red blood cells than liver. Sorry for the confusion? But nevertheless, as you can use PC to extract DNA from whole blood without special measures to cope with erythrocyes, I would just give it a try!
