Nonradioactive overgo probes - (Feb/08/2007 )

Hello all,

I have a genomic BAC library that I would like to screen with overgo probes (36 to 40-mers created from two oligos with complementary sequence). Easy enough, but the problem is that our lab is not commissioned for radioactive materials and our PI would like to keep it that way.

Does anyone know how I can end-fill these probes with some type of hapten (fluorescein, digoxigenin or biotin)?

there are protocols for biotinylating oligos, it's particularly easy if you end-label them prior to annealing but there are several variations and I have had excellent luck with the kit from Pierce
