which -20 or -80 is more less degradation of cell lysates - (Feb/07/2007 )

After lysis cells using RIPA buffer that contains PMSF and PIC,

I will try to store immediately the lysates at -80 or at -20 after adding equal volum of 2xSDS sample buffer

which storage condition is the best way for p-AKT WB ?

I think for short-time storage (up to 2 weeks) it should may no difference, although it is not recommended to freeze lysates with sample buffer as performance in gel electrophoresis is often worse than with freshly added sample buffer; long-term storage should be done at -80°C or liquid nitrogen

Just curious. What is RIBA and PIC? I know PMSF is protease inhibitor.

Actually, I thought it wouldnt be a really good idea to keep your lysate in -20 or -80 as thawing might degrade your protein or even change the structure.

For me, I usually kept it in 4Celcius and only kept for a day before running SDS PAGE.

-80 is better for storing protien extracts. We make lots of aliquots so that one doesnt have to do multiple freeze thaws.
