increased/decreased expression in western blot - (Feb/01/2007 )

Hi,
I got quantification results from my western blotting. But, I do not know if there's a really increased or decreased expression according to what I have from the quantification of my blot. How can I get an accurate result for this? I wonder if anybody can help me with this problem? Thanks

it is difficult to interpret "expression" of protein form Wb; we better talk about "abundancy" of a protein of interest in your cell system to a certain time point detected by a certain antibody;

for quantification of blots you need several blots of distinct experimets (at least n=3) to say sth about differences in significant lower or high adundancy compared to control

Its quite difficult to quantify expression using WB.

A crude method would be to load different vol. of the sample and have also dif. vol. of control. Do a WB.

After the WB, use a program to quantify the intensity of each band making sure the bands are not completely saturated. Using the intensity, one could kind of quantify the expression levels. Once again, this is a crude method.
