How design a linker - (Jan/12/2007 )

Hi,

I need to design a linker to close a vector but I never do it before. I need a linker with HindIII on one side and pstI site on other side the side. I would like also put a multiple site of cloning inside this linker. Is someone else have any background to construct this kind of linker.

What I did was to replace the MCS of pbluescript with a linker of aropund 30 bp. I ordered primers that annealed together live the overhangs complimentary to thos of the linearized vector. 5 new multiple cloning sites in my linker. I order them but phosphorylated and unphosphorylated.... both worked fine without need of further phosphorilation.
