To prepare fresh or not for sucrose gradient - Could it be stored longer? (Dec/25/2006 )

Hi,

I was wondering, within what time-frame do I have to use my layered sucrose gradient before it's not gradient anymore (e.g. due to diffusion)? Could the sucrose gradient be stored more than a day (or what's the maximum hours you've stored your gradient before using it)?

i always use the gradients fresh. if you allow them to sit they will even out.

I have not tried, but store it at 4C should slow its diffusion. You can add some dye to the lower phase and see how it diffuses as time goes.

when pouring new gradients, add some phenol red to one of the two sucrose solutions; so you can visualize the gradient and get a feeling how long it will be stable by macroscopic inspection
