CNS injury - (Dec/12/2006 )

Dear friends.,

Do you have any ideas any model for CNS injury?I have search it for papers mentioning animal model especially rat,but still not satisfied me,there is still have a way out! help me

what sort of injury are u looking for?

One could damage the brain by opening the skull and using a pressure associated piston to deliver the injury or one could cut the optic nerve, which is widely accepted model for traumatic injury and since optic nerve is a part of the CNS, all the more better.

Scolix, thanks for your response. I have a problem to introduce the injury inside to the brain, which not opening the skull as mentioned, but depth to the basal ganglia. i haven"t found any animal model for injury without opening the skull. Is there any method to approach the injury from basal ganglia??

Ok, now u have to be more specific. Which region(s) do u want to injure.

if its substantia nigra, then u could chemically change it.

u could try initiating ischemia, then globus pallidus is most severely affected.

Thanks, But what happened if the injury occured at eg putamen or caudate nucleus?, how do i introduce the injury into these area?What condition it may mimics in human condition?

Cerebral ischemia wants to mimic human stroke, and there are some models without opening the skull. You have to occlude an artery, and this occlusion can be transient or permanent. Ischemia models are usually called MCAO (middle cerebral artery occlusion), and there are several ways to do that: for example occluding transiently by inserting and removing it after some minutes a nylon filament or thread (this requires practice) through the common carotid artery or by using clamping clips.

I think that idea would be for stroke induced model and could be posibble as CNS injury and am glad to take note that. Anyway my plan to mimics Diffuse Brain injury in Human spesifically in basal ganglia portion. The idea is there are some cases in ward that contributed very little cases approximately 2-3% involving Injury to Basal ganglia, so called diffuse brain injury grade3. BUT, the question is : i dont have any ideas how to mimics this kind of injury in animal model. There are certain chemically induced but still these chemical were commonly used for Parkinson DISEASED INDUCED Animal model. How about mechanically induced trauma? do i need to follow animal model or otherwise?

Ok, I see. Isn´t anything publish about what you want? Nobody has done this injury before? If there isn´t a established model, you could set up one wink.gif

I think that if you want to publish that, referees will ask you for a "serious" model (I mean, established -by you or other people-, reproducible, ...).

Pumuki, thanks for idea and link! To built a model need some 'serious' efforts too which reproducible (as you said! which is i am agree), sensitivy and spesificity of the model, better than existing model. I dont think DAI(diffuse Axonal Injury) dont have any animla models,still searching for good model. Have you been published any animal model before?

No I haven´t. I´m still too young (2nd year PhD student) rolleyes.gif
