PDGFR labelling - (Dec/12/2006 )

I'm working with PDGF induced oligodendroglioma cells, I'm trying to label my fixed cells for p -PDGFR-beta labeling but somehow I'm getting signals all over from cytoplasm and not restricted to the cell membrane periphery. Any suggestions are welcome.
Same when i tried to label ePDGFR it is giving signals inside the cell which it should not be. I'm not using triton in this case........Please help me in catching the fault as I'm doing ICC for the first time sad.gif

PDGF-R can suffer internalization in vesicles by different stimulus. Maybe that's the cytoplasmic signal.

Use specific antibodies, preferentualy mABs; do staining on ice (if cells are not fixed).
