Does set up PCR mix days prior the amplification affects the results? - (Dec/07/2006 )

Does anyone did some test like this?!

If I set up my PCR mix with everything (dNTP, primers, Taq...) except the DNA template, and leave it at the freezer (-20oC), and like a week or two later I add my template and put at the machine...

Does this week of "waiting" affect the reaction?

I have only leave it overnight without problems, but if you have to make lots of reaction in no much time you could use pcr beads.
Just need water and template. Amersham have it and they are really good no need freezer, but I recommend store in a dry place at vacumm.

I always keep a stock PCR mix (including Taq) in the freezer with everything but my template and primers. I've used it weeks after making it. You should be good! smile.gif
