Is my RNA dead? - freezing cells after adding RNA lysis buffer (Dec/01/2006 )

Hi!

I am new to this forum, but hoping it is the right one to pose my question in, which I haven't found the answer to anywhere:

I added RNA lysis buffer to my cultured cells and froze the mixture (-70). According to the Ambion protocol, one should not do this, but I am wondering why, and, is my RNA ruined now? Anybody know anything abt this?

DO not worry. RNA lysis buffer lyses the cells and not RNA. So your RNA is safe at -80. You can follow the further protocol and get good RNA. All the best. Try searching for "RNA lysis buffer" and see what the search results say..
