Is multiple freezing-thawing harmful to cDNA? - (Nov/28/2006 )

I rather prefer to freeze aliquots of cDNA in TE (pH 8) rather than cooling in a refrigerator; may multiple freezing-thawing harmful to cDNA? what do you think?

Ideally, you do not want to freeze-thaw cDNA too many times, so the best option is to prepare small aliquots of it and keep them at -20 to minimize the number of freeze-thaw cycles. Having said that, I've had to freeze-thaw cDNA several (5/6) times and I haven't found it to be damaged...

but what actually speaks against multiple freeze-thawing? Can there really sth damaged?

I might be wrong, but my idea is that freeze-thawing might break up the cDNA in fragments...
