Protein analysis - (Nov/15/2006 )

I was wondering if anyone could help me. I would like to analysis both soluble protein and membrane protein from leaf tissue. I have seen some papers where they use sucrose gradients and I was wondering if this is the best way to go about this.

what kind of analysis do you want to do?

if you just want to show how many proteins and what size and pI you can run 2d electrophoresis.

if just size then sds-page.

sucrose gradients may help you separate groups of protein based on buoyancy but will not give you much information.

I want to see if i can find where a certain protein im looking for is accumulating.

the sucrose gradient can be used to separate organelles then you can assay for your protein but you may want to use immunohistochemistry or immunocytochemistry to localize your protein.
