spleenocyte count for hybridoma production - (Oct/30/2006 )

Hi,
I make hybridomas using mouse spleenocytes and myeloma cells ( one spleen to about 50,000,000 myeloma cells).
I will soon be creating a hybridoma using rat spleen.
How can I count the rat spleenocytes (regular camera count, or use a dye?)?
How may rat spleenocytes to how many myeloma cells?

hi milli,
the ratio between splenocytes to myeloma cells is a surface phenomenon.
in general i used to make ratio 1:5k-10K (myeloma vs splenocytes) coz splenocytes are much smaller in size.

so with rat liver also same dilution should work, there is no hard n fast rule behind the ratio.
