DEPC treatment, EDTA? - (Oct/25/2006 )

Hello,

can you please tell me if it is possible to treat EDTA with DEPC? Is there a list anywhere were you can look up which substances can be treatet and which not? I always read that some substances like tris cannot be treatet, but I am not good enough at chemistry to deside for myself what else cannot be tratet.

as EDTA contains amine residue it can't be DEPC treated but should be prepared with pretreated water

I think DEPC only reacts with the primary and secondary amines, but not with the tertiary amines. EDTA has only the tertiary amines in its structure. So I think it would be safe to treat EDTA solution with 0.1% DEPC before autoclaving.

I read: "Make up Tris-Cl (pH 7.6) from a fresh bottle in autoclaved, DEPC-treated H2O. Prepare NaCl and EDTA in 0.1% DEPC in H2O. Store for at least 12 hours at 37°C and autoclave the mixture for 15 minutes at 15 psi (1.05 kg/cm2) on liquid cycle. "

It seems that their protocol is excerpted from the newest "Molecular Cloning" by Sambrook, although Maniatis is no longer in the author list.
