Anyone use phusion HF DNA polymerase? - urgent!!!! (Oct/24/2006 )

Hi everybody:

Anyone use phusion(Finnzyme)? How about its effect in amplfying long and GC rich chain?!

Thanks!

Hi,

I've used phusion, but I guess I haven't tested it out to much. I was able to easily PCR amplify 2-3Kb inserts accurately. As far as GC content the products were all only around 55-60% GC so haven't stressed the system too much.

Cheers,

Scott

phusion comes with a buffer specific for gc-rich or 'difficult' templates...I would try it

I have had excellent luck with that enzyme

Scott and aimikins:

Thanks for your kind reply!

I would try it, BTW, I wonder that whether it shows better than pfu in fidelity according to your experience?

in my hands it, I got way more amplification with phusion than with pfu. Btw you can also order get it under the name iProof Polymerase from Bio-rad. I think it's a bit cheaper

Hi Kersten:

Do you mean the Fidelity and proceeding ability of iProof Polymerase from bio-rad is nearly identical with phusion?!

Thanks in advance!

i never compared the two directly, but somehow, if I read the product description, they sound very much the same. I would assume, that at the end they are the same even.

iProof always worked pretty fine for in my hands
