Standard oligos T7 and T3 - (Oct/23/2006 )

can somebody tell me a bit more aout the T7 and T3 standard primers? t7 promoter, what is that?

The T7 promoter is bound by the T7 polymerase. This is a very active and high fidelity phage polymerase

T3 and T7 primers binds to known regions in a wide variety of cloning vectors and serve as universal sites for sequencing a cloned product. They are helpfull if your product is too large for sequencing with a normal Fwd and Rev pair or primers, i.e. sequencing is easier if the template is shorter. You can use an internal primer in your product and T3 and T7 primers for sequencing towards extremes.

In other words,
T7 and T3 can act as promoters, or just hybridization site for sequencing primer, or both, which depends on the purpose they are placed in the vector in the first place, e.g. cloning vector or expression vector.
