ELISA Incubation time? - (Oct/23/2006 )

What will be the consequence if the primary and secondary antibody of captured ELISA are incubated for 2 days in 4 degree C separately? Will they denature?

Also, for the process of blocking? What is the difference between using non-fat dry milk and BSA?

Q1: Ab should not be denaturized

Q2: milk contains various proteins but predominantly casein; BSA is bovine serum albumin; if there is no existing or well working protocol, one has to test which blocking solution will be the best; depends very much on Ab and antigens
