Ethanol precipitation vs Clean up kit? - which will yield more (Oct/19/2006 )

Pages: Previous 1 2

I'm with kits all the way -- quck, reliable, reproducible, controllable...

Which kit is the best for ppt DNA?

Well, I used DNazol (not really a kit though), have to use EtOH to ppt DNA.

But isnt it if the DNA sample is not dry enough, EtOH will inhibit PCR reaction right? correct me if I am wrong. Cheers

That's correct. You should remove as much EtOH as possible by pipetting and then let your sample dry for some minutes to be sure (that's the way I dry my samples, always worked fine).

Thanks for reassuring. tongue.gif
EtOH is pretty hard to be removed. Is there any other faster or more efficient method to remove EtOH?

How about under laminar flow?

And is it really significant affect on PCR from EtOH? Anyone got any effects from EtOH in your PCR results?

I place my samples on a heating block at 90 degrees for 1 minute to evaporate residual ethanol. Apart from inhibiting enzymes, ethanol makes your sample very difficult to load onto a gel....samples with ethanol will float out of the well.

in our lab, we use a modulspin31 to vaporize ethanol

Usually, I use GFX PCR and DNA Gel Band Purification Kit from Amersham (currently GE Healthcare)

It works fine and I never had any problems
It's fast, reliable and reproducible.
