hai all
i am interested in ligating restriction digested inserts of genomic dna into a vector (8 kb). what should be the insert vector ratio for ligation?

which one is 8 KB your vector? how about insert?

but that is just the quick rule of the thumb. Vary the insert:vector molar ratio according to the difficulty of the ligation.

my vector is 8 kb. i want to insert random genomic fragments into it aftr. cutting both the vector and the genomic DNA with a restriction enzyme.
so the insert size is unknown here. will the 1: 3 ratio work well in this situation too?
