Unknown receptor identification... - (Oct/03/2006 )

Hey there

I was just wondering if anybody out there has any ideas on how to identify the unknown receptor that a known ligand binds to? We really know very little about the interaction (ie. it might even be internalised or it could be binding on the cell surface). If anybody has any suggestions I'd love to hear them.

after usual cell lysis, bind the proteins to biotin, setup an affinity chromatography system supported with avidin, add the ligand...

by the way, what is your ligand?

other possibility -depending on your ligand- is to immobilize it and to pour with matrix-carrying ligand an affinity column to fish the unknown receptor from lysates; lots of methods to immobilize are available

as kosmodrom suggest, u can immobilize your ligand (it depends) to agarose beads then fish your protein which should be labelled..
