dNTP calculation - (Sep/25/2006 )

If I mix 100ul of 100uM each dNTP (ATP, CTP, GTP and TTP) to make 400ul(4x100ul) , which is "dNTP mix concentration"?


1) 400uM dNTP mix 400ul
2) 100uM dNTP mix 400ul
3) 25uM dNTP mix 400ul

which one is right?

I would say you have 25mM.

that is the concentration of each type of nucleotide, which is probably the more important thing to the polymerase, rather then the total concentration of all dNTP

I agree; 25 µM of each nucleotide (but overall 100 µM of nucleotides - but this is irrelevant)
