loading control for human serum - (Sep/15/2006 )

hello again,

Could somebody recommend me a loading control for human serum Western Blot?
I can´t find anything anywhere. I´ve asked to a lot of technical supports but nobody knows.

I meant what kind of loading control do you want it to be? Do you want to show how good is your gel separation, or positive/negative control for WB?

I would like to test if I am loading always the same amount of protein. I mean, to ensure that the differences I may see between lanes are real, and not due to a wrong loading, so I need a protein whose concentrations in serum doesn´t change, so their bands must be equal in all lanes, isn´t it?.
Sorry, I´m still very inexperienced with WB and at the moment I´m alone on this sad.gif

You can assay for serum protein concentrations and based on the readings load the same amount of protein. Bradford assay takes a few minutes to complete.

Maybe someone else can comment on if there is a protein that has a level among different individuals that is close enough to serve as an internal control.
