Iam a PhD student working on the gene expression of host macrophage in disease and after different treatment. I am not know very much about whats precaustion should be taken in primer disign for RT-PCR and which free available software good for its.
I hope you peoples tell me whats precaution shuld be taken for its and software also.

I would advice you to have a look at the website of your RT-PCR supplier. At least AB gives you guidelines on the primer design
