Counting methods - dinoflagellates (Sep/02/2006 )

can anyone give some methods that how can i count the cells?

I grow the cells in the 500ml conical flask and i want to know the number of cells in the 500ml flask,

Dont know much about dinoflagellates but can suggest u use a haemocytometer or any flow cytometer.

Try counting chambers eg. Abbe-Zeiss counting cell chamber. They look like similar to normal microscope slides and have a big cover slip. They have a defined volume (made from the slip) were you then apply your well mixed sample. You count cells in the a part of the grids (the surface is segmented by a grid) under a microscope and can calculate total cell number. It is normally used for blood cells etc.

There is a book "Manual on harmful marine microalgae", one of the chapter has mentioned how to do the cell counting of dinoflagellates. This is a really good book for people working on dinoflagellate research
