Fixation for eGFP imaging? - what fixation should I use? (Sep/01/2006 )

I know that fixation of transiently transfected cells w/eGFP can weaken or abolish the fluorescence of eGFP.

I cannot seem to find a general standard for the fixation of cells w/eGFP so that I can go image them. They need to be fixed.

The localization of the eGFP will be cytoplasmic, no specific cellular location.

I fix cells with 4% PFA and GFP still fluoresces

try also 4% icecold methanol especially if you like to do immunocytochemistry afterwards

-The Bearer-

even 4% formaldehye works nicely. No loss of fluorescence.

Also the mounting media can make a difference in fluorescence.
