gateway system - how it works? (Aug/30/2006 )

HI I tead about gateway system but Iam not sure how it work.
first you have to put you gene if interest in a entry vector.
then you mixe this vector for several other ve tor or just one other vector?

u need to clone ur gene or whatever piece of DNA into the entry vector. then with the help of a simple enzymatic reaction, u can transfer to a expression vector. Usually one gets more than 90% correct clones using this method.

hm .. with LR reaction only 90% correct clones? i didn't use it much, but, up tp now i always got the right clone, therefore i was thinking about not sequencing the expressionclone anymore. would you then recommend to keep sequencing the expression clone?

It is more like 99.9% Sequencing is most probably not nesecary but you can do a restriction digestion

90% right clones with the insert. I dont sequence it, just verify by restriciton digest.

okay, I agree on that... if insert is present, sequence is right
