RT-PCR primer design - (Aug/28/2006 )

Hi all,

I hate to ask the stupid question, but i am kind of confused now. When designing primers for RT-PCR, should I follow the sequence of mRNA or cDNA?

If you do it manually follow the sense strand of DNA for one primer (forward) and antisense for another (reverse). If you use software, feed it with sense (antisense wont do any harm either).
