From yeast to mamall cells - principles and methods online? (Aug/22/2006 )

I am changing the field from yeast cell biology to mammals cell and cell culture (research on carcinome).
As I am new in the area I would like to ask you for tipps and normal protocols used for cloning (for ex retroviral cloning, transfection and so on). Is there any book online or similar?

Invitrogen has a manual for lentiviral system, which one could use to get an idea for cloning and transfection and virus production.
