plasmid construction - (Aug/22/2006 )

Hello,
I would like to construct the plasmid for RNAi expermint. First, I have to change antibiotic marker from hygromycin (in pSilent1) to benomyl (obtained from pBT6). Please suggest me in the detail of how to change or critcal point that I should concern it. I don't have any experence in this experiment.
Also, pBT6 does not have sequence show in database. We know that it derives from pUC12.

U need a vector map of pBT6 to know the location of benomyl. if its derived from pUC12, look at its map.

try to find some common sites to exchange the antibiotic resistance gene.
