Optical Adhesive Covers - bubbles (Aug/18/2006 )

Before I run a 384 well plate, I put on the optical cover and spin the plate at 1000 RPM for 1 minute. I noticed that ater the spin, I have a bubble in nearly every well. Tapping the plate does not get rid of it.
So I have been removing the optical adhesive cover and taking a 10 ul pipet tip and touching the bubbles to pop them. (One tip per well). Then i put on a new cover and run the PCR. The standard deviations of three replicates are usually good, except when I am unable to pop the bubble. I am using SYBR Green. Anyone else noticed this?

We used to get bubbles in 96 well plate format. When cover is on and you have ensured there is a good seal turn plate upside and tap a few times against palm of hand or if only a few wells flick bottom of well using your thumb nail and then centrifuge. This usually worked.
