problem with SDS-PAGE - (Aug/09/2006 )

Hi,

I run SDS-PAGE many times, but every time the marker runs very weired. I use Benchmarker from sigma. On the top of gel, the marker runs as widely as the normal. However, at the bottom the marker runs very narrowly, like a line. The closer to the bottom, the narrower the marker is. I don't know why. Does anybody have the same problem?

you may have high salt content in the sample in the lane next to your marker.

20mM Tris, 300mM NaCl and 10mM Imidazole is in my sample.

dialysis or gel filtration to 50 mM NaCl will eliminate your problem.

i think, but I'm not 100% sure, that you can add the same amount of salts in the marker and in the lane beside your sample to solve the problem.

no need to dialyse if you add NaCl to your marker it should resolve the problem,

i have done this in the past up to 0.8M NaCl
