Total RNA extraction from ethanol preserved samples? - (Aug/08/2006 )

Hi ...

First experience with RNA....could we get RNA from ethanol preserved samples?????
is it we always have to use fresh samples fro RNA extraction???/

Thankyou.....

I have a related question- is there a way to isolate Total RNA from archival tissue that is fixed and paraffin embedded like brain tumors? -Thanks

hi...

kindly give reply if someone knows.......

thankyou

hi
at which temperature were they stored?
what kind of sample are they ? cells, tissues?...
what's the purpose of the extraction? (blot is ok, but may be tough for qRT-PCR)

Hi...

Prawns stored at room temperature in 70%alcohol.....
is it possible to extract RNA...and go on with first strand cDNA synthesis and RT-PCR..
Thankyou....

well... how long were they stored...
Were they also just putted in alcohol?...

They were just directly put into 70% alcohol a week back and stored at room temperature....

well i think for RNAextraction, ethanol is not a bad thing.
But for quality of the prep, i guess the problem is how much RNAses can act before the ethanol enter the middle of fruit...
I think the RNA extraction is fully feasible, but i would separate prawn in few layers, cause i'm supposing there may be quality differences between the center and periphera.
les me know the result.
