ELISA for frozen fat tissue - (Jan/31/2002 )

I would like to quantify cytokines in fat tissues with ELISA and Western. The following procedure described elsewhere for fresh fat tissue could be applied to frozen one? Thank you for your attention.
"...fat tissue was extracted in 1 ml of a solution of 0.1M TRIS HCl, pH 8.0, and 6 M guanidine hydrochloride and this suspension was shaken continuously at room temperature overnight. The suspension was centrifuged at 10 000 × g for 10 minutes and the supernatant fat tissue extract was collected... the total protein concentrations in the fat tissue extracts were measured..."

I am sure it will work for frozen tissue. Freezing should not harm the cytokines, rather preserve them. The tissue will thaw quickly at room temperature.
