Could we freeze the 0.25 trypsin EDTA and thaw it, use it?
we try to aliquot the a bottle of 0.25% trypsin EDTA in 10ml tubes, could we freeze them, and everytime thaw one tube and use it. will it reduce the activity of trypsin by freezing it?

We aliquot trypsin in 10ml tubes and use one at a time. As long as one has them in 4C after thawing, it works fine.

After thawing, we store our 100 ml trypsin bottle at 4C for a couple of months without any problem.

That is exactly what I do in my lab and it has so far worked out well for us. I aliquot 10-12 ml per tube and freeze them down and when I'm ready to use them I just thaw out what I need. I don't know if there are any bad effects to doing this. The trypsin seems to be working just fine on all of my cells.

In my labs we encourage researchers to aliquot all tissue culture products including FCS, Glutamine, Trypsin, Pen/Strep, NEAA, Hepes solution, Sodium Bicarbonate etc.
I was always told that the aliquoting of 100ml/500ml solutions was good laboratory practice and meant that :-

a) The product was not Frozen and thawed many times over many months.

i freeze the volume that will be required eg 10 ml and thaw one aliquot of FCS, pen-strep, L-glut and Trypsin EDTA as needed - never had any problems

we use same way as pcrman. Storeat 4° for monthes.
But defrost, aliquot and storage is ok for trypsin.

we aliquot in single-use portions, store at -20, and thaw just before use

we are paranoid about contamination, we study skin cells and common skin pathogens...very very easy to contaminate the samples. so, I guess it depends on what you're doing, as well?
