SiRNA and GFP transfection MCF7 cells - (Aug/01/2006 )

Hi

I succesfully transfected MCF7 Cells with GFP however when I used the same exact conditions optimized for GFP to transfect SiRNA the trasfection didnt work at all. Is it normal that the conditions that works for GFP is not necessiraly work for SiRNA ?

How did you do the transfection, i.e. reagent, ratio?

It doesnt sound like a transfection related problem. Did you design the siRNA yourself? How did cells look after trasfection? Did they look sick to you?

also, what % EGFP positive cells do you think it was accoding to your measurement/estimation? 30-50% or more?
