Problem amplifying 8kb bacterial genomic DNA - (Jul/31/2006 )

Hey guys. I´m trying to amplificate a 8kbp fragment for a bacterial genomic DNA and dosen´t function. I was probing Elongase, Taq GO and some mix of enzymes and nothing. The primers are ok but the DNA its a little complicate to extract but function with more little amplificates (2kbp).
Well, some sugestions please?

What is the GC content of the organism? Can you test the primers by using them to amplify shorter fragments?

Hi, thanks for your request, the GC contet is 58,7. sadly I can´t probe the primers with small fragments. What you can suggest me?
