Immunohistochemistry without perfusion - (Jul/31/2006 )

Help!
Is perfusion absolutely necessary for carrying out immunohistochemistry on rat brain sections?
Can I remove the brain (without perfusion), separate left and right hemispheres and then embedd in paraffin?

No it isn't...You could do small frozen sections. But if you are going to embed in paraffin, I'd perfuse for the fixation. Handling brain tissue that is not fixed, is worse than handling Jello.


Is there a reason you are opting not to fix in that manner?

Better perfuse to drain out the blood else u could have autoflurescence and high background after Immunohistochemistry.

Also fixed brain is easy to handle without much damage.
